I'd suspect the shoes first off

Are they either very new or old (>500 miles done in them)?

The skin on your feet will have softened after time off so it will take a couple of weeks for it to harden up again. I recently started again after not being able to run for nine months and got some blisters; I just ignored them and ran through it and the feet are OK now (but this was in shoes I know and trust - I use Saucony 10s and replace every 500 miles; if you over- or under-pronate though you need a different style of shoe from that).

Other things to try: Vaseline on the feet, two pairs of socks. But honestly my first thought is "wrong shoes".
