I think it's less of a problem these days; but the rule of thumb used to be
to keep at least 10% of disk space free to allow the operating system to shuffle stuff around when it needed to. Again, fragmentation used to be a problem; but OS X handles that these days (but might conceivable be hamstrung by a nearly full disk). Anyway, it might be a good idea to get another hard drive and move your music library to it.
Are you up to date with OS X and iTunes? Have you tried running Disk Utility?
