Take a rondvaart (tour) in a boat from just outside Central Station. Don't do the dinner and drinks one as the food and drinks are s**t. Do it in daylight.
Rijksmuseum for sure. Allow plenty of time. Maritime museum if that interests you. Van Gogh museum if you like that s**t (I hate his work). Walk around red light district of course.
Eat at the Caf? Bern on Nieuwmarkt - amazing fondue and steak but again book in advance as it is very popular and very small. Drink in Nieuwmarktplein as it is far more Dutch than Leidseplein and Rembrandtplein which are too touristy. Avoid any bars or restaurants on the Dam like the plague as they are tourist traps. Shop on the Rokin and Kalverstraat.
Nightlife - try to go to one of the very cool lounge bars, or the Supperclub, rather than the usual tourist haunts. Best behaviour to get in though and doesn't hurt if you have some women in your group.
If you have five days then also see somewhere else which is more Dutch. Haarlem is a short train journey away, Delft and Utrecht are also within an hour. Can give more advice about Delft if you fancy that (I lived there for 8 years). Or you can come to the Hague (where I live now) and visit the Mauritshuis (complete with Girl with the Pearl earring), Escher museum (fantastic) and the Binnenhof (houses of parliament).
Loads to see and do.
