Stupid question about credit cards

I have just got an American Express card. Can I make as many payments as I wish, whenever I want without getting penalised?

I basically want the card to get the rewards, pay it off as soon as I use it. So buy something for ?50, pay it immediately with my debit card.

I relinquished the reward thing, by going for a card I can transfer money over
